Each contestant brings a one million yen participation fee. The last person to stay in the room without laughing wins. The winner is awarded a 10 million yen prize and bragging rights as the funniest comedian.

After participating to a previous show where they have to bet or be challenged to any kind of competitions, the losing members of the Gaki no Tsukai are selected to spend 24 hours in a setting which they have no knowledge of the theme nor the course of the events. They are exposed to various absurd and crazy situations nonstop for 24 hours and must refrain from laughing. If they do so, hang-men appear and hit the rule breakers with a weapon (the weapon varies every year).

Kazunari Ninomiya × Masayasu Wakabayashi (of Audrey) team up for the first time as co-hosts to present a brand-new type of survival game variety show, “Secret NG House.” In this game, Ninomiya and Wakabayashi act as the Game Masters, setting hidden rules — “Secret NGs” (absolutely forbidden actions). If a player performs any of these NG actions three times, they are immediately eliminated. The players must use the warning sounds that go off when an NG action occurs as clues to deduce what the forbidden actions are. Those who figure it out can then set traps to make their unsuspecting rivals commit the NG actions, knocking each other out in the process! After driving back the Kanto-based Tojin Alliance, the Osaka hangure group Battle Family, led by Kenji Tsukino (Ryohei Abe), stands victorious. Yet the powerful Kanto Sanrenwakai, which controls the Tojin Alliance, continues to seek revenge as part of its planned expansion into Kansai. One night, Battle Family members are drinking at their usual hangout—a bar owned by Yuya (Kaname Endo)—when Kenji is confronted by an old acquaintance, gangster Fujiyoshi (Honkon). Tensions flare, and Kenji and Yuya end up beating Fujiyoshi to death. Unbeknownst to them, Fujiyoshi was a sworn brother of Hanashiro (Hitoshi Ozawa), the wakagashira of the Fujimasa-gumi, where Kugisaki belongs. To settle the score, Kugisaki summons Kenji and orders him to kill Yuya. At the same time, the Tojin Alliance begins moving into Kansai, hunting down the Battle Family. With enemies closing in from all sides, a new and deadly conflict begins.
