Shipwreck survivors found on the presumably uninhabited Infant Island leads to a scientific expedition that discovers a surviving native population along with the Shobijin, tiny twin fairy priestesses of the island's mythical deity called Mothra. After the fairies are kidnapped by an exploitative businessman named Clark Nelson, Mothra sets out to rescue them.

Kanbei Hamidashi aka Tsukada Kambei was originally a direct retainer and head of a large group but could not fit into the organization and spent his days drinking and fighting. This is the first film in the third installment of the adventure film for boys that depicts the battle between a thief who calls himself "the secret envoy of the Demon Lord" and private investigator Hideo Sayama. In the Toei, the roles are twenty faces and Kogo (Twenty Faces) and Akechi Kogo, with a content that seems to be strongly conscious of the "boys' Detectives" series of Edogawa Rampo, which was later made into a movie.
